▣ 초  록

Single molecule localization microscopy (SMLM) has significantly advanced our ability to visualize subcellular structures with superior spatial resolution down to 20 nm. Awarded the 2014 Nobel Prize in Chemistry, this super-resolution technique has facilitated the exploration of new nanoscale structures within cells, enhancing our understanding of their associated physiological functions. Despite its success in single-cell imaging, extending SMLM to more complex biological samples such as tissues and small animals still presents significant challenges. This seminar begins with an introduction to the foundational concepts of SMLM and how it surpasses the resolution limit of conventional fluorescence microscopy. I will then introduce our innovative adaptation, oblique plane SMLM (obSTORM), which is designed for a diverse range of biological samples, from tissues to whole small animals. Finally, I will present our lab’s recent efforts to enhance imaging performance of obSTORM through several convergent approaches, including developing a high refractive-index SMLM imaging buffer, advancing software for improved point spread function modeling, and applying deep learning techniques to effectively remove background fluorescence. By integrating these methods, obSTORM will significantly broaden the applications of SMLM, offering new opportunities for deep tissue investigation and expanding its impact on the fields of biology and medicine.
